New Technology Speeds Up Preparation of DNA Libraries

DNA libraries have become critical tools for the diagnosis and treatment of diseases such as cancer. DNA libraries can take on many forms. In this article, a “DNA library” refers to DNA fragments such as circulating cell-free patient DNA (cfDNA) that are attached to specialized adapters. They enable amplification of the library using Polymerase Chain Reaction. Such libraries prepared from an individual patient can be utilized to understand the genetic basis of underlying physical conditions such as cancer.

This patent portfolio addresses the need for high complexity DNA libraries from biological samples such as cfDNA. It provides novel methods and reagents specifically for targeted DNA sequencing using next-generation DNA sequencing (NGS). The intended applications include DNA sequence-based liquid biopsies. One key feature of the patented invention is that it improves the efficiency of cloning and the physical representation of targeted DNA fragments in NGS. The target regions for the technology are easily “programmable” using simple oligonucleotide probe sets and hybridization-based capture in optimized conditions that are described with the method. In a further embodiment of the patented technology, each clone is uniquely labeled. This results in NGS datasets that are amenable to quantitative analysis. The technology is ideally suited to the customized, patient-specific detection of minimal residual disease in individuals undergoing treatment for cancer.
